First of all, players of the first will instantly notice that Dante looks different. Some people say he doesn't look as cool, but there's a reason for that. This game, as previously stated, is headed by Tsuyoshi Tanaka. Tanaka states that "What I think is cool and what Mikami thinks is cool are two totaly different things."
Dante is given a more rugged look to show that he's a much more experienced devil hunter, and though I'm not sure of this, I believe quite a lot of time has passed between numbers 1 and 2. Dante now has a more Solid Snake tough-guy attitude than a Duke Nukem cool/tough guy attitude.
Dante is now equipped with a new arsenal of moves. He can walk on walls, as nearly all of us know, shoot upside down in the air, and even use melee attacks in midair, like flying kicks. He can also flip during a double jump, and execute long dives. Dante's coolest move, though, is his ability to shoot enemies behind him. Even cooler is his ability to shoot one enemy behind him with one pistol, and peg the one in front of him with the other! I have seen pictures of this, and it looks sweet.
Enemies will die in new ways, too, like bursting into polygon pieces or just turning into dust. The use of Red orbs will be enchanced as well, but Tanaka doesn't explain how.
Now, as many of you know, there is a rumored female character. This is not a myth, there IS a new character, playble from the start. Her name is Lucia, the "protector". (If any of you have played Lunar 2, this Lucia is very different from the one you know.)
If you're unsure of what she looks like, here's the best description I can give. She has orange hair, cut in a style so that half of it covers her left eye. She wears denim jeans, sort of, and a similar top covered by a white semi-cloak. She might have demon powers, but I'm not sure. You can count on it, though. She does not use guns, from anything I've seen, but she carries two swords in a crescent shape. She also is skilled in martial arts.
Tanaka explains that "Many people complained in the first game, because they wanted to play as Trish, and use a heroine. We agree, so we're giving them what they want." This isn't merely a new character with different moves placed in the same quest. Lucia can be chosen from the beginning, and she will have a very different storyline. Expect some different weapons, too.
According to Tanaka, each level is estimated to be nine times larger than those from the original, though they have only started work on 3 levels. One of the newest elements to these levels, however, is the non-linear pathways. For example, in the level that looks like a dilapidated town, you can either travel through the streets, or climb up the wall and jump from roof to roof! This is just one example of the types of originality in these levels.
Don't expect getting rankings to be easy, you'll need to search high and low for monsters to kill and items to find. If you liked Secret Missions, they're back too, but they'll have a different approach, though this is uncertain.
Also, you won't have to constantly go back to previous areas. The puzzles in the newest game will feature a more straightforward approach, so that you don't complain about returning to the drawbridge, or whatever.

The other seems to be a heliport, like in Metal Gear Solid 2, which probably has a boss. There is also the aforementioned dilapidated town.
The visual detail of the levels has been enchanced as well. The graphics look a lot sharper than the original.
Now, a lot of people complained about the first DMC. Some said it was too easy, or too hard. Well, DMC2 will have an engine similar to that of Jak and Daxter, which changes the difficulty depending on your progress in a level. Cool, huh?
Another annoyance was the camera work. Sometimes, the bosses were so large that some other objects got in the way of your view. However, Tanaka is trying his best to make sure the camera work is a lot better, and judging from the pictures in front of me, they're damn good. The view is still fixed, but it's a good idea, since you don't have to always spend time re-adjusting the camera yourself while the enemies put holes in you.
Also, there are less objects, like tall pillars or trees, to obstruct the camera's view during battles, so the camera can easily pull back or change views when needed. Dante still seems to shoot offscreen at enemies on occasion, but this will probably be fixed, and isn't always that annoying.
